چکیده

Gene silencing by feeding double-stranded (dsRNA) holds promise as a novel pest management strategy. Nonetheless, degradation of dsRNA in the environment and within insect gut, well inefficient systemic delivery are major limitations to applying this Branched amphiphilic peptide capsules (BAPCs) complexed with have been used successfully target genes outside inside gut epithelium upon ingestion. This suggests that BAPCs can protect from shuttle it across epithelium. In study, our objectives were 1) Determine whether on BAPC-dsRNA complexes targeting putative peritrophin gene P. japonica would result suppression synthesis, 2) gain insight into cellular uptake mechanisms transport larval midgut japonica. Our results suggest readily taken up epithelium, treatment tissue endocytosis inhibitors effectively suppresses intracellular transport. Further, assessment expression BAPC- fed beetles demonstrated significant downregulation mRNA levels relative control and/or alone. increase efficacy knockdown alone adults. To knowledge, is first report nanoparticle-mediated through .

Branched amphiphilic peptide capsules: cellular uptake and retention of encapsulated solutes.

Branched amphiphilic peptide capsules (BAPCs) are peptide nano-spheres comprised of equimolar proportions of two branched peptide sequences bis(FLIVI)-K-KKKK and bis(FLIVIGSII)-K-KKKK that self-assemble to form bilayer delimited capsules. In two recent publications we described the lipid analogous characteristics of our BAPCs, examined their initial assembly, mode of fusion, solute encapsulatio...

How does RNA editing affect dsRNA-mediated gene silencing?

In general, double-stranded RNA (dsRNA)-binding proteins (dsRBPs) are not sequence-specific. A dsRNA molecule in a cell will interact with any dsRBP it comes in contact with, suggesting that different dsRNA-mediated pathways intersect and affect each other. This paper analyzes evidence that the ADAR RNA editing enzymes, which act on dsRNA, affect dsRNA-mediated gene silencing pathways. Examples...

Silencing of Aphid Genes by dsRNA Feeding from Plants

BACKGROUND RNA interference (RNAi) is a valuable reverse genetics tool to study gene function in various organisms, including hemipteran insects such as aphids. Previous work has shown that RNAi-mediated knockdown of pea aphid (Acyrthosiphon pisum) genes can be achieved through direct injection of double-stranded RNA (dsRNA) or small-interfering RNAs (siRNA) into the pea aphid hemolymph or by f...

Molecular basis of double-stranded RNA-protein interactions: structure of a dsRNA-binding domain complexed with dsRNA.

Protein interactions with double-stranded RNA (dsRNA) are critical for many cell processes; however, in contrast to protein-dsDNA interactions, surprisingly little is known about the molecular basis of protein-dsRNA interactions. A large and diverse class of proteins that bind dsRNA do so by utilizing an approximately 70 amino acid motif referred to as the dsRNA-binding domain (dsRBD). We have ...

On the Role of RNA Amplification in dsRNA-Triggered Gene Silencing

We have investigated the role of trigger RNA amplification during RNA interference (RNAi) in Caenorhabditis elegans. Analysis of small interfering RNAs (siRNAs) produced during RNAi in C. elegans revealed a substantial fraction that cannot derive directly from input dsRNA. Instead, a population of siRNAs (termed secondary siRNAs) appeared to derive from the action of a cellular RNA-directed RNA...
